The primary role will include providing technical research services directly supporting the research protocols, working in partnership with research staff members in the conduct of biomedical research to include:

• Provide support for large scale mutagenesis screens, including generating fish lines, conducting relevant complementation testing and map-crossing, managing screen related data, and PCR-based fine mapping (marker analysis) to narrow down and/or isolate the mutant region/lesion.                                           

• Perform technical procedures related to Aquatic species (e.g. obtaining blood and other samples, obtain zebrafish sperm for cryopreservation and in vitro fertilization, facilitate animal breeding and embryo collection, identify pathogens in zebrafish (P. neutrophilia and Mycobacterium species) by PCR and DNA sequence analysis).Assist post-doc and collaborators with research projects. Teach post-doc zebrafish reproductive physiology and care.

• Assist with lab-based technical resource / instrument procurement and manage NICHD Program equipment usage.Assist with all aspects of zebrafish related experimental design, including drafting SOPs and protocols for zebrafish related tasks; statistical analysis of data; generation of publication quality images, graphs and figures; scientific writing for publication.

• Utilize a variety of online zebrafish genome databases for gene searches, candidate gene identification for mutation analysis, genomic DNA sequence analysis for PCR primers, mRNA / Protein sequence analysis, and CRISPR target analysis / generation.

• Technical and bench top duties including embryo cleaning, staging and fixation, mutant and transgenic identification and propagation, microinjection of DNA and RNA into zebrafish embryos, gDNA / RNA isolation, mRNA / cDNA synthesis, PCR, gel electrophoresis, in situ hybridization, molecular cloning, whole mount antibody staining, fluorescent microscopy,  western blotting, and CRISPR / Cas9 mutant zebrafish generation (all steps from gRNA target generation to locating genomic lesion in F2 mutants).

• Help screen established mutant zebrafish lines using fluorescent microscopy, PCR / Restriction Enzyme Digest analysis, KASP genotyping assays on BioRad CFX, and Fluorescent PCR analysis on ABI 3130xl. Manage international and domestic shipment of zebrafish embryos and adults following Federal and NIH guidelines.

• Troubleshoot aquatic support equipment and provides input and action on cause and remedy. 

 

Applicants must possess: B.S. in Biological Sciences; at least 3 years in a laboratory research environment including basic molecular biology tasks. Aquatic Species experience in a research is highly desired.
