  1. understand the architecture of rare pediatric genetic disease,

  2. investigate the cellular mechanisms underlying disease processes, and

  3. identify novel therapeutic paradigms for inherited disorders.

Additionally, candidates will have the opportunity to investigate unique pediatric cohorts; use cell-based, zebrafish, or mouse models; and employ cutting-edge molecular and imaging technologies in a state-of-the-art facility (https://www.feinberg.northwestern.edu/sites/simpson-querrey/ ).

  • Carry out phenotypic and genotypic screening of mutant and transgenic lines.

  • Perform basic molecular biology manipulations such as DNA/RNA/protein purification and quantification, expression vector construction, morpholino injection and CRISPR/Cas9 gene editing.

  • Express mutant proteins in vitro to examine effects such as protein stability, cell/nuclear morphology and the cell cycle.

  • Perform embryo/cell imaging experiments including fixed and live-cell confocal microscopy.

  • Analyze human genetic variants for their effects on specific protein domains and cellular functions.

  • Interact with a team of trainees, technical staff, and faculty to optimize laboratory techniques and oversee the day-to-day management of multiple projects, performing advanced experimental design, optimization and interpretation.
